The AmpliScribe™ T7 Aminoallyl-RNA Transcription Kit enables high-yield synthesis of aminoallyl-labeled RNA (AA-RNA). The kit incorporates 5-(3-aminoallyl)-UTP (AA-UTP) into the RNA transcript. Since AA-UTP is incorporated into RNA with the same efficiency as UTP, high yields of AA-RNA are obtained. The AA-RNA product can then be treated with amine-reactive biotinylation reagents or fluorescent dyes to obtain biotin-labeled or fluorescently labeled RNA, respectively. Reaction conditi*** h***e been optimized to c***istently generate uniformly labeled AA-RNA that, upon conjugation to a fluorescent dye or biotin compound, yields high signal intensity. This method for making nonradioactively labeled RNA is more efficient and less expensive than direct incorporation of fluorescent- or biotin-labeled NTPs into RNA during in vitro transcription.
